What Is CJC-1295?

CJC-1295 is a modified analog of growth-hormone-releasing hormone, built on the same GHRH(1-29) backbone as Sermorelin but with additional amino acid substitutions for protease resistance, plus a Drug Affinity Complex (DAC) — a maleimidopropionic acid linker that reacts with a free thiol group on circulating albumin, covalently attaching the peptide to it. That albumin binding is what gives CJC-1295 its defining property: an estimated half-life of 5.8–8.1 days, versus the few minutes native GHRH or Sermorelin last in circulation.

This is also a common source of confusion in the research-peptide space: what's sold as "CJC-1295 without DAC" is a related but distinct, shorter-acting peptide (often called Mod GRF 1-29) that lacks the albumin-binding modification entirely — it is not simply CJC-1295 minus one feature, but a different compound with a very different duration of action.

Clinical & Preclinical Trial Data

CJC-1295 has been tested in humans more extensively than most compounds referenced on this site. The pivotal study (Teichman et al., Journal of Clinical Endocrinology & Metabolism, 2006; 65 healthy adult subjects) found a single injection produced a dose-dependent GH increase of 2–10× baseline sustained for at least 6 days, and an IGF-1 increase of 1.5–3× baseline sustained for 9–11 days — consistent with the compound's estimated 5.8–8.1-day half-life. With repeated dosing, mean IGF-1 remained above baseline for up to 28 days, and the compound was well tolerated with no serious adverse events reported.

A separate study (Ionescu & Frohman, Journal of Clinical Endocrinology & Metabolism, 2006) examined healthy men receiving a single CJC-1295 injection with overnight GH sampling before and one week after. GH secretion increased with preserved pulsatility — the frequency and magnitude of GH pulses were unaltered — but trough (basal) GH levels rose 7.5-fold, contributing to a 46% increase in mean GH secretion and a 45% increase in IGF-1.
