How CJC-1295 Was Discovered
CJC-1295 was developed in the early-to-mid 2000s by ConjuChem Biotechnologies, a Montreal-based biotech that specialized in a technology it called Drug Affinity Complex (DAC) — a chemical linker capable of attaching a peptide drug covalently to circulating albumin, the most abundant protein in blood plasma. Applied to GHRH(1-29), the same fragment underlying Sermorelin, the DAC modification solved GHRH's core practical limitation: its native half-life of only a few minutes. Albumin-bound CJC-1295 instead persists in circulation for an estimated 5.8–8.1 days.
The pivotal human data came from a study of 65 healthy adult subjects (Teichman et al., Journal of Clinical Endocrinology & Metabolism, 2006), which found a single injection sustained elevated growth hormone and IGF-1 levels for over a week — a dramatic improvement over unmodified GHRH.
ConjuChem's broader ambitions for the compound didn't survive contact with a later-stage trial. The company was running a multicentre, randomized, placebo-controlled Phase II study of CJC-1295 in 192 participants with HIV-associated lipodystrophy and visceral obesity when, in July 2006, a study participant at a trial site in Argentina died a few hours after an injection. The attending physician's assessment was that the most likely explanation was asymptomatic coronary artery disease unrelated to the study drug, but ConjuChem suspended the trial as a precaution. CJC-1295 never advanced to Phase 3 or FDA approval, and remains an investigational research compound rather than an approved pharmaceutical.
