What Is FOXO4-DRI?

FOXO4-DRI is a D-retro-inverso peptide — a synthetic peptide built from mirror-image (D-) amino acids in reverse sequence, a design technique that confers resistance to normal enzymatic degradation. It's engineered to disrupt the FOXO4-p53 interaction that senescent cells depend on to evade apoptosis (programmed cell death), competitively blocking that interaction so p53 is freed to trigger cell death specifically in senescent cells while sparing healthy ones.

Research Focus: A Senolytic Approach

Clinical & Preclinical Trial Data

All data is preclinical, from a single landmark study (Baar et al., Cell, 2017). Using naturally aged mice, a chemotherapy-induced senescence model, and a fast-aging XPF-ERCC1-deficient mouse strain, intraperitoneal FOXO4-DRI (5mg/kg, three times weekly) reduced senescence markers (p21, γ-H2AX), improved kidney function markers, restored fur density in areas of alopecia, improved exercise tolerance, and accelerated recovery from doxorubicin chemotherapy, relative to controls. No human clinical trial has been conducted for any indication.
