How FOXO4-DRI Was Discovered
FOXO4-DRI's design starts from a specific biological observation: senescent (“zombie”) cells rely on an interaction between the FOXO4 protein and p53 to evade apoptosis (programmed cell death) that would otherwise clear them. Researchers engineered FOXO4-DRI to competitively disrupt that interaction, freeing p53 to trigger cell death specifically in senescent cells while sparing healthy ones.
It is built as a D-retro-inverso peptide — a synthetic peptide constructed from mirror-image (D-) amino acids in reverse sequence, a design technique that confers resistance to normal enzymatic degradation without changing the molecule's side-chain topology.
What FOXO4-DRI Has Been Studied For
The landmark 2017 study (Baar et al., Cell) used naturally aged mice, a chemotherapy-induced senescence model, and a fast-aging mouse strain, finding that intraperitoneal FOXO4-DRI reduced senescence markers, improved kidney function markers, and restored fur density in areas of hair loss. This established FOXO4-DRI as an early proof of concept for the senolytic research strategy: selectively clearing damaged, senescent cells rather than supporting the function of cells that remain healthy.
