How FOXO4-DRI Was Discovered

FOXO4-DRI's design starts from a specific biological observation: senescent (“zombie”) cells rely on an interaction between the FOXO4 protein and p53 to evade apoptosis (programmed cell death) that would otherwise clear them. The landmark 2017 study (Baar et al., Cell) used naturally aged mice, a chemotherapy-induced senescence model, and a fast-aging mouse strain, finding that intraperitoneal FOXO4-DRI reduced senescence markers, improved kidney function markers, and restored fur density in areas of hair loss. This established FOXO4-DRI as an early proof of concept for the senolytic research strategy: selectively clearing damaged, senescent cells rather than supporting the function of cells that remain healthy.
