What Is PT-141?
PT-141 (bremelanotide) is a cyclic heptapeptide developed by Palatin Technologies as a structural analog of Melanotan II, itself a synthetic analog of alpha-melanocyte-stimulating hormone (α-MSH). Unlike Melanotan II, which was originally studied for skin pigmentation, PT-141 was developed specifically around its activity at melanocortin receptors relevant to sexual function — primarily MC4R, expressed in the hypothalamus and limbic system. Its proposed mechanism is central (via the nervous system) rather than the peripheral vasodilation mechanism used by drugs like sildenafil.

Clinical & Preclinical Trial Data
Two identical Phase 3, randomized, double-blind, placebo-controlled trials known as RECONNECT (NCT02333071 and NCT02338960) enrolled premenopausal women with hypoactive sexual desire disorder (HSDD), testing bremelanotide 1.75mg self-administered subcutaneously as needed over 24 weeks. Both trials found statistically significant improvements in sexual desire and related distress compared to placebo, published in Obstetrics & Gynecology in 2019 (Kingsberg et al., 2019). Based on this data, the FDA approved bremelanotide in June 2019 under the brand name Vyleesi for acquired, generalized HSDD in premenopausal women — the first FDA-approved melanocortin receptor agonist for any indication.
